I have Comcast Business Internet -- Downloads up to 50Mbps, uploads up to 10Mbps. Is that fast enough to use instead of ISDN for voice over sessions? Is there a website where I can test my ISDN-ready connectivity?

It should be, although you may want to pick up Sourceconnect or similar software to allow your clients to remotely record your session.
ISDN is essentially dead technology and is being phased out be major telecommunication providers. It is expensive to order and maintain for anyone using it.
Some clients demand ISDN for their projects, which is unfortunate. If you have a client like this, it's most likely easier and cheaper to rent a studio with an active ISDN connection than it is to install and maintain ISDN service. Just work the studio rental into your fee. If the client balks at the price, offer to connect with them via Skype or phone for direction and offer to lower your price.
